A practical reference on AEDG: what it is, how it behaves, what the literature reports, and where the honest uncertainties sit.

Animal and clinical reports appear mainly in Russian-language journals from the 1990s and 2000s, covering endpoints such as melatonin rhythm, lifespan in aged rodents, and retinal function. Many of these papers involve small groups, lack blinding or placebo comparison, and are difficult to retrieve through indexed databases. Review articles published in English generally summarise the claims without reanalysing the underlying data. Because no large randomised trial exists, the clinical importance of these reported effects stays unresolved and is properly described as an open question.
No national medicines regulator has approved epitalon as a therapeutic product. It is generally distributed as a research chemical, and in some jurisdictions selling peptides for human consumption without approval is restricted or prohibited. Certificates of analysis accompanying commercial material vary in which tests are performed, and independent verification of identity and purity is uncommon. Statements about anti-ageing or disease-prevention benefits on vendor pages are marketing claims rather than regulatory findings, a distinction that shapes how the compound is discussed in scientific and popular sources alike.
The parent extract epithalamin was characterised as a low-molecular-weight fraction of pineal tissue rather than a single defined chemical entity. Researchers fractionated it and tested successive fragments for activity, a screening approach typical of peptide discovery work in that era. Epitalon emerged from that process as one of the shorter sequences of interest. Because the original extract was never fully resolved into individual components, claims about which constituent drives a given effect rest on inference. This distinction matters when reading older reports that attribute extract observations to the tetrapeptide itself.
Epitalon is a synthetic tetrapeptide with the residue sequence alanine-glutamate-aspartate-glycine, commonly abbreviated AEDG. Its monoisotopic mass is approximately 390.35 daltons, and it is usually supplied as a lyophilised trifluoroacetate or acetate salt. The compound was derived from a pineal gland extract called epithalamin, a heterogeneous preparation investigated in the former Soviet Union. Researchers associated with the Saint Petersburg Institute of Bioregulation and Gerontology described the tetrapeptide as a constituent fragment of that extract. Commercial material is offered as a laboratory reagent rather than as a finished pharmaceutical product.

Dissolution is usually performed in water or a suitable aqueous buffer, and the resulting liquid is divided into single-use portions before freezing. Freeze-thaw cycling is a recognised source of loss for short peptides, since each cycle can encourage aggregation or adsorption onto container walls. Working solutions are generally kept cold and used within a short window, although published stability data specific to epitalon are thin. Containers should be marked with concentration and date, and solutions examined for cloudiness before use.
Identity is normally established by reversed-phase high-performance liquid chromatography combined with mass spectrometry, a pairing that separates components and confirms molecular mass at once. Purity is quoted as a percentage from the chromatogram, and figures above ninety-five percent are a frequent commercial specification. Amino acid analysis or sequencing supplies further confirmation when required. Because many short peptides behave similarly under chromatography, retention time alone does not establish sequence; the mass measurement is what separates one tetrapeptide from another, and certificates should report both.
Short peptides such as AEDG are normally supplied as a freeze-dried solid and are kept dry, cold, and shielded from light. Holding at minus twenty degrees Celsius is common for the long term, while a refrigerator suffices for brief periods before use. The material takes up moisture to some degree, so containers should stay sealed and be allowed to reach room temperature before opening, which limits condensation on the contents. Repeated warming and cooling of a single container is discouraged because it admits water and can lower the amount of intact peptide.
Interest in epitalon is usually discussed within the broader field of short peptide bioregulators, a category that includes other synthetic di-, tri-, and tetrapeptides studied by the same research group. These compounds share a common rationale: that small fragments of tissue-derived proteins can retain biological activity and can be produced reproducibly. The category as a whole remains outside mainstream pharmacological consensus, and epitalon specifically has a limited presence in independent, non-Russian research literature, which shapes how its evidence base is described.
Epitalon is a synthetic tetrapeptide with the amino acid sequence alanine-glutamate-aspartate-glycine, abbreviated Ala-Glu-Asp-Gly or AEDG. It was developed by the Russian researcher Vladimir Khavinson and colleagues during work on peptide bioregulators derived from the pineal gland. The compound is short enough to be produced by standard solid-phase peptide synthesis and is typically handled as a lyophilized white powder. Its small size distinguishes it from larger pineal peptides such as epithalamin, a complex extract from which the tetrapeptide was conceptually derived.
The four residues give epitalon a molecular formula of C14H22N4O9 and a molecular weight near 390.35 daltons. The presence of two acidic residues, glutamate and aspartate, makes the free peptide strongly acidic, while the alanine and glycine ends provide neutral, nonpolar character. This combination produces a molecule with substantial water solubility. Because there are no cysteine, methionine, or tryptophan residues, the peptide lacks the most common oxidation-sensitive side chains, which simplifies handling compared with many longer peptides.
The most widely cited claim is that epitalon activates telomerase and thereby extends telomere length. Supporting evidence comes largely from cultured human cells, where treatment was associated with increased telomerase activity and delayed replicative senescence. Telomerase activation is a biologically consequential effect, since the enzyme is largely silenced in most somatic cells. However, the route by which a short peptide would reach and act on the enzyme's regulatory machinery has not been established. Independent replication in human trials is scarce, so the link between cell-culture observations and whole-body aging remains an open question.
Human data are limited to small studies, often without the randomization, blinding, or control groups expected in contemporary clinical research. Reported outcomes have included changes in melatonin levels, immune markers, and subjective measures, but sample sizes were generally too small to support firm conclusions. Some reviews treat the peptide as promising while noting methodological weaknesses; others question whether the observed effects are specific. The compound is frequently discussed in longevity-focused communities, where enthusiasm often outpaces the published evidence. Separating established findings from speculation is therefore important when reading summaries of this research.
Epitalon emerged from research conducted in Saint Petersburg by Vladimir Khavinson and colleagues, who studied short peptides as potential regulators of aging. The work built on epithalamin, a pineal gland extract reported to influence neuroendocrine function. Epitalon was designed as a synthetic counterpart with a defined sequence, allowing reproducible experiments that extracts could not support. Early publications described effects on melatonin rhythms and lifespan in animal models. These findings circulated mainly in Russian-language journals during the 1990s, which limited their visibility among English-speaking researchers.
The monofunctional monomer, often referred to as a chain stopper, controls and limits the polymerization of bifunctional monomers because the growing polymer yields chain ends devoid of functional groups and therefore incapable of further reaction.
July–September 2024 Welsh Labour leadership election: Welsh Labour officials agree a timeline for the election, with a new leader to be in place on 14 September. Figures produced by the Institute for Fiscal Studies suggest the UK government would need to spend an extra £3bn to implement a 5.5% pay rise for teaching and NHS staff. 21 July It is confirmed that the bodies of Briton Sarah Packwood and her Canadian husband Brett Clibbery, who disappeared on 18 June while sailing their eco-friendly yacht across the Atlantic Ocean, were found on Sable Island near Nova Scotia on 12 July. Six people are killed in a crash between a car and a motorbike on the A61 road near Barnsley. 22 July – BT is fined £17.5m for a "catastrophic failure" of its emergency call handling service which led to thousands of 999 calls not being connected. 23 July Following a trial at Woolwich Crown Court, radical preacher Anjem Choudhary is convicted of directing the banned Islamist organisation Al-Muhajiroun. The Home Office confirms that the contract for the Bibby Stockholm, the vessel used to house asylum seekers, will not be renewed after January 2025. A report from the National Audit Office warns that the UK government may be forced to discourage people from travelling by train between Birmingham and Manchester after the Birmingham–Manchester leg of HS2 was scrapped. This is because HS2 trains will still continue on to Manchester, but will have fewer seats than the present rolling stock.
== Structure == Spiroligomer molecules can be synthesized in any direction, and between any pair of bis-amino acids. Spiroligomer diketopiperazines can be created between either end of a bis-amino acid. Spiroligomer molecules are known to be conformationally rigid, due to the fused-ring backbone.

Although every organ system secretes and responds to hormones (including the brain, lungs, heart, intestine, skin, and the kidneys), the clinical specialty of endocrinology focuses primarily on the endocrine organs, meaning the organs whose primary function is hormone secretion. These organs include the pituitary, thyroid, adrenal glands, ovaries, testes, and pancreas. An endocrinologist is a physician who specializes in treating disorders of the endocrine system, such as diabetes, hyperthyroidism, and many others (see list of diseases).
Within the international relations literature, international organizations facilitate cooperation between states by reducing transaction costs, providing information, making commitments more credible, establishing focal points for coordination, facilitating the principle of reciprocity, extending the shadow of the future, and enabling interlinkages of issues, which raises the cost of noncompliance. States may comply with the decisions of international organizations, even when they do not want to, for rational cost-benefit calculations (to reap concrete rewards of future cooperation and avoid punishment) and normative reasons (social learning and socialization). International organizations vary in terms of: inclusive or exclusive membership; broad or narrow scope; equal or unequal control by members; precision and flexibility of rules; hard or soft obligations; and delegation of power to the international organization. Other examples of international organizations include the UN General Assembly, the World Trade Organization, the African Development Bank, the UN Economic and Social Council, the UN Security Council, the Asian Development Bank, the International Bank for Reconstruction and Development, the International Monetary Fund, the International Finance Corporation, the Inter-American Development Bank, and the United Nations Environment Programme.
Serotonin is a major neurotransmitter in the nervous system and contributes to formation of new neurons (neurogenesis), formation of new connections between neurons (synaptogenesis), remodelling of synapses, and survival and migration of neurons, processes that are necessary for a developing brain and some also necessary for learning in the adult brain. 45% of ASD individuals have been found to have increased blood serotonin levels. Abnormalities in the serotonin transporter have also been found in ASD individuals. It has been hypothesized that increased activity of serotonin in the developing brain may facilitate the onset of ASD, with an association found in six out of eight studies between the use of selective serotonin reuptake inhibitors (SSRIs) by the pregnant mother and the development of ASD in the child exposed to SSRI in the antenatal environment. The study could not definitively conclude SSRIs caused the increased risk for ASD due to the biases found in those studies, and the authors called for more definitive, better conducted studies. Confounding by indication has since then been shown to be likely. However, it is also hypothesized that SSRIs may help reduce symptoms of ASD and even positively affect brain development in some ASD patients.
== Applications == The nanoDSF technology was used to confirm on-target binding of BI-3231 to HSD17B13 and to elucidate its uncompetitive mode of inhibition with regards to NAD+. NanoDSF was used to compare the thermal stability of a matched set of anti-CD20 antibodies representing a range of variants. The results revealed a spectrum of activities. NanoDSF has also been applied in drug repurposing and anticancer research. In a large-scale screen of FDA-approved molecules, the method was used to identify several dozen novel microtubule-targeting agents (MTAs), including clinically used drugs such as aprepitant, disulfiram, and toremifene, by directly demonstrating their interaction with tubulin and their effects on microtubule polymerization.

=== Route of administration === The most common route of administration at a methadone clinic is in a racemic oral solution, though in Germany, only the R enantiomer (the L optical isomer) has traditionally been used, as it is responsible for most of the desired opioid effects. The single-isomer form is becoming less common due to the higher production costs. Methadone is available in traditional pills, sublingual tablets, and two different formulations designed for the person to drink. Drinkable forms include ready-to-dispense liquid (sold in the United States as Methadose), and Diskets (known on the street as "wafers" or "biscuits") tablets which are dispersible in water for oral administration, used similarly to Alka-Seltzer. The liquid form is the most common as it allows for smaller dose changes. Methadone is almost as effective when administered orally as by injection. Oral medication is usually preferable because it offers safety, and simplicity and represents a step away from injection-based drug abuse in those recovering from addiction. U.S. federal regulations require the oral form in addiction treatment programs. Injecting methadone pills can cause collapsed veins, bruising, swelling, and possibly other harmful effects. Methadone pills often contain talc that, when injected, produces a swarm of tiny solid particles in the blood, causing numerous minor blood clots.
Severe side effects are rare and occur mostly in those with poor immune function. Its use in people with HIV/AIDS should be done with care. It is not recommended during pregnancy; however, the few times it has been given during pregnancy no problems resulted. The vaccine is available either by itself or along with the MMR vaccine, in a version known as the MMRV vaccine. It is made from weakened virus. A live attenuated varicella vaccine, the Oka strain, was developed by Michiaki Takahashi and his colleagues in Japan in the early 1970s. American vaccinologist Maurice Hilleman's team developed a chickenpox vaccine in the United States in 1981, based on the "Oka strain" of the varicella virus. The chickenpox vaccine first became commercially available in 1984. It was first licensed for use in the US by Merck, under the brand name Varivax, in 1995. It is on the World Health Organization's List of Essential Medicines.
== Release and promotion == BioShock's first public showing was at E3 2006. After a fan petition calling for a special edition of the game reached 5,000 signatures, 2K Games announced a limited edition of the game, featuring a Big Daddy figurine, making-of materials, and the game's soundtrack. Before the special edition was released, the proposed soundtrack CD was replaced with The Rapture EP. BioShock was released on August 21, 2007, in North America, and August 24 in Europe and Australia for Windows and Xbox 360 platforms. The demo released the week before release was so popular the demand crashed Xbox Live's servers. The Windows release shipped with SecuROM copy protection that required activation from 2K Games' servers over the Internet; the unavailability of these servers was reported as the reason for the cancellation of the game's midnight release in Australia. Complaints from players caused 2K Games to raise the simultaneous installation limit for the game from two to five copies; the activation limit was removed after the release of the game, though other copy protection methods remained. Levine admitted that their initial approach to the activation process was malformed, harming their reputation during the launch period. The initial release of the game also cropped the top and bottom of the field of view in order to fit widescreen monitors, resulting in less vertical view instead of more horizontal view compared to 4:3 monitors.

Small studies with human participants have been reported, chiefly in Russian-language journals, but they are limited in size and design. No large randomised controlled trial with published results is available.
The most reproducible biochemical observation comes from cultured cells, where the peptide was reported to raise telomerase activity. Whether this translates into measurable health effects in animals or humans is not established.
It is offered as a research chemical for laboratory use, which places it outside the approval pathways applied to medicines. Buyers should expect documentation to differ between suppliers.
It is based on epithalamin, a peptide-rich preparation obtained from pineal gland tissue and studied in the former Soviet Union. Researchers later identified the tetrapeptide AEDG as a constituent fragment of that material. Modern supplies are produced by chemical synthesis rather than tissue extraction.
